Thématiques de l’équipe
Les axes de recherche de l’équipe sont majoritairement dirigés vers la mise au point de nouvelles techniques d’analyses miniaturisées. Il s’agit notamment de développer des méthodes à l’échelle micro voire nanométrique dans un but de pré-concentration, de séparation, de caractérisation et de quantification de molécules d’intérêt biologique telles que des peptides, protéines ou anticorps. Ces méthodes permettent également l’étude d’interactions entre les molécules d’intérêt et le milieu biologique qui les contient. Le panel de techniques mis en place est utilisé pour (i) le diagnostic ou le suivi thérapeutique via l’analyse de biomarqueurs ; (ii) le contrôle pharmaceutique et la caractérisation de protéines thérapeutiques ; (iii) la caractérisation de nano-objets , notamment les vésicules extracellulaires, les agrégats d’anticorps et les nanoparticules; (iv) la recherche ou la validation de nouveaux biomarqueurs de pathologies.
Le laboratoire est actuellement impliqué dans 6 thématiques majeures :
- Le développement d’approches innovantes pour l’isolement et la caractérisation de vésicules extracellulaires, ainsi que la détection de biomarqueurs exosomaux.
- Le développement de méthodes pour la détection et la caractérisation d’anticorps monoclonaux dénaturés et petits agrégats.
- Le développement de méthodes électrocinétiques et de nanochromatographie liquide haute performance pour l’amélioration de la séparation et de la détection de protéines/peptides.
- La synthèse de supports solides ou monolithes pour le traitement d’échantillons biologiques : enrichissement, immunocapture, digestion enzymatique…
- La conception de dispositifs en gouttes et nouveaux concepts instrumentaux (tels que la LEGO-CE, la magnéto-CE, ou les détections multimodales) pour des outils de diagnostic miniaturisés.
- Le développement d’outils de diagnostic et de méthodes de criblage de molécules à visée thérapeutique contre les maladies neurodégénératives, le diabète de type 2 ou certaines anomalies de glycosylation.
