The INPACT team develops nano- and microparticulate systems for drug delivery and diagnostics based on physico-chemical and pathophysiological concepts, with a particular focus on the treatment of inflammatory diseases and cancer. To assess the effectiveness of these systems, the team has established a wide range of biological and pharmacological models.

In recent years, their objectives have included the design of microparticulate systems capable of remaining at the site of action (lungs, inner ear, arteries), the development of functionalised nanomedicines able to target immune or cancer cells following intravenous administration (via aptamer or polysaccharide functionalisation), the delivery of coding (mRNA) and non-coding nucleic acids (antimiRs, siRNAs), and the creation of nanoparticle systems for imaging and photoacoustics.

Recent key results include:

(i) Functionalisation of liposomes with aptamers (anti-CD44 and anti-nucleolin) enhances tumour accumulation and the efficacy of interfering RNAs or HSP90 inhibitors.
(ii) Pickering emulsions based on lipiodol and stabilised by poly(lactide-co-glycolide) nanoparticles loaded with oxaliplatin show high efficacy in hepatocellular carcinoma treatment by chemoembolisation.
(iii) A new process for the synthesis of dexamethasone palmitate nanoparticles improves the reduction of inflammatory symptoms in a murine model of rheumatoid arthritis. The efficacy of an anti-miR155 antagomir delivered via PEGylated liposomes and an anti-TNFα siRNA delivered via cationic phosphorus dendrimers was also demonstrated in the same model.
(iv) Improved targeting of pro-inflammatory macrophages following intratracheal administration of hyaluronic acid-coated liposomes.
(v) Development of hybrid systems (liposome-hydrogel) to prevent injury and inflammation during cochlear implantation.
(vi) Design of BODIPY-based nanoparticles with high photoacoustic efficiency.
